Principe
HiPure FFPE Nucleic Acid Kit, designed for rapid and efficient extraction of both DNA and RNA from formalin-fixed, paraffin-embedded (FFPE) tissue samples:
Key Features:
- Silica Gel Column Technology: Utilizes a reliable and streamlined approach, eliminating the need for harsh chemicals like phenol-chloroform.
- Fast Processing: Extracts DNA/RNA in just 20 minutes, excluding digestion time.
- Sample Compatibility: Works effectively with various FFPE tissue sections.
Processus d'extraction:
- Deparaffinization: Xylene removes paraffin wax, a common component used to preserve tissues.
- Lysis and Digestion: A special solution (lysate) and an enzyme (protease) break down cells and proteins, releasing nucleic acids (DNA and RNA) into the solution.
- De-Crosslinking: Heating at 90°C reverses the effects of formalin fixation, allowing easier access to nucleic acids.
- Binding to Silica Membrane: After adjusting conditions with ethanol, the sample is transferred to a column. DNA/RNA binds to the silica membrane, while proteins pass through.
- Washing: Buffers remove impurities like proteins and salts from the captured nucleic acids.
- Elution: A low-salt buffer elutes the purified DNA/RNA, making it ready for downstream applications.
Applications:
- RAP (réaction en chaîne par polymérase)
- Southern Blot
- Viral nucleic acid detection
- Other analyses requiring high-quality DNA or RNA
Avantages
The HiPure FFPE Nucleic Acid Kit offers a superior solution for extracting DNA and RNA from formalin-fixed, paraffin-embedded (FFPE) tissue samples.
Sécurité:
- No Xylene Contact: Eliminates the need for hazardous xylene, minimizing exposure to harmful chemicals.
Speed:
- Fast Processing: Extracts DNA/RNA in just 20 minutes (excluding digestion time), allowing you to process multiple samples within 2 heures.
- No Overnight Incubation: Streamlines the workflow by eliminating the need for lengthy overnight incubation steps.
Haute efficacité:
- Enhanced PCR Sensitivity: Effectively removes formaldehyde modifications from DNA, significantly improving the sensitivity of downstream PCR (réaction en chaîne par polymérase) analyses.

Stockage et stabilité
- Protéinase K: Keep it cool in the refrigerator (2-8°C) upon arrival for maximum effectiveness.
- Short-Term Storage (jusqu'à 12 semaines): Proteinase K can last at room temperature (15-25°C) for up to 12 weeks without issues.
- Everything Else: Store other kit components at room temperature (15-25°C). They are stable for at least 18 mois dans ces conditions.
- Important: If storing the entire kit cold, remember to warm up the buffers before use. All buffers should be at room temperature for optimal results.
