This product provides an easy-to-use workflow for selective isolation of bacterial DNA from samples that are intrinsically rich in host DNA, such as body fluids or swabs. The method is specific for the identification of intact bacteria so it prevents false results due to nucleic acids from dead bacteria. The Kit allows isolation of enriched bacterial DNA suitable for a variety of applications, including qPCR and whole metagenome or 16S rRNA gene sequencing.

Načelo
Ovaj proizvod se temelji na pročišćavanju kolonom silicijevog dioksida. This product efficiently depletes human and animal host DNA and yields enriched bacterial DNA. An optimized combination of mechanical and chemical lysis allows efficient disruption of bacterial cells. Target DNA is adsorbed on the membrane, dok se protein ne adsorbira i uklanja se filtracijom. Nakon pranja proteina i drugih nečistoća, nukleinska kiselina je konačno eluirana s puferom s malo soli (10mm Tris, pH 9,0, 0.5mm EDTA).

Skladištenje i stabilnost
DNase I and Proteinase K should be stored at 2–8°C upon arrival. Međutim, kratkotrajno skladištenje (do 12 tjedni) na sobnoj temperaturi (15–25°C) ne utječe na njihovu izvedbu. Preostale komponente kita mogu se čuvati na suhom na sobnoj temperaturi (15–25°C) i stabilni su najmanje 18 mjeseci pod ovim uvjetima. Cijeli komplet može se čuvati na 2–8°C, ali u ovom slučaju pufere treba ponovno otopiti prije upotrebe. Provjerite jesu li svi puferi na sobnoj temperaturi kada se koriste.
