Prinsip
- HiPure Plasmid Procedure:
- Based on alkaline lysis of bacterial cells followed by adsorption of DNA onto silica in the presence of high salt.
- Utilizes a unique silica membrane that replaces glass or silica slurries for plasmid DNA minipreps.
- Procedure Steps:
- Preparation and Clearing of Bacterial Lysate:
- The alkaline method is used to prepare and clear bacterial lysate.
- DNA Binding:
- Supernatant transferred to a column to bind DNA to the silica membrane.
- Washing and Elution:
- Proteins and other impurities washed away.
- Nucleic acid was finally eluted with low-salt buffer (10mm Tris, pH9.0, 0.5mm EDTA).
- Preparation and Clearing of Bacterial Lysate:

Penyimpanan dan Stabilitas
Komponen kit dapat disimpan kering pada suhu kamar (15-25°C) dan setidaknya stabil 18 bulan dalam kondisi ini. Jika ada endapan yang terbentuk di buffer, hangatkan pada suhu 37℃ hingga larut. Setelah penambahan RNase A, Buffer P1 stabil untuk 6 bulan bila disimpan pada suhu 2-8°C.
