HiPure Circulating DNA Midi Kit, designed for efficient purification of short, cell-free DNA (cfDNA) molecules from human biofluids for clinical use:
- Target:
- Tumor-derived cfDNA fragments and mRNAs in blood
- Fetal nucleic acids in maternal blood
- Sample Compatibility:
- Human plasma
- Human serum
- Human urine
- Samples can be fresh or frozen (once only)
- Focus on Short DNA: The kit isolates short nucleic acids (<1000bp) commonly found in biofluids.
- Efficient Purification: The protocol allows for effective recovery of cfDNA from various biofluids.
- Clinical Applications: The purified cfDNA is suitable for use in in vitro diagnostic (IVD) tests approved for clinical purposes.
Benefits:
- Optimized for Short cfDNA: Specifically designed to capture short DNA fragments relevant for biomarker discovery and liquid biopsies.
- Broad Sample Compatibility: Works effectively with various human biofluids, offering flexibility in sample selection.
- Larger Scale Purification: The Midi Kit format allows for processing of larger sample volumes compared to Mini Kits, potentially increasing cfDNA yield.
- Clinical Utility: Purified DNA can be directly used in approved clinical IVD tests.
This kit empowers researchers and clinical laboratories studying circulating DNA for applications in oncology, non-invasive prenatal testing (NIPT), and liquid biopsy analyses, ultimately aiding in patient diagnosis and management.

Principle
This product offers a fast and reliable method for purifying DNA using silica column chromatography.
- Lysis and Digestion: The sample is treated with a solution and enzyme to break open cells and degrade proteins, releasing DNA.
- Binding to Silica: The lysate is transferred to a column. The silica membrane in the column captures DNA molecules, while impurities pass through.
- Washing: Wash buffers to remove unbound proteins and cellular debris.
- Elution: A low-salt buffer elutes the purified DNA from the silica.
This method provides a valuable tool for researchers working in various fields that require purified DNA for analysis.
Advantages
- High yield – most optimal process, free DNA (>50bp) can be obtained to the maximum extent
- High concentration – low elution volume, ensuring high nucleic acid concentration
- High purity – low alcohol binding method, completely removing inhibitor and protein pollution
- High recovery – DNA can be recovered at the level of PG by silica gel column purification

Storage and Stability
- Keep it cool: Store Proteinase K and Carrier RNA in the fridge (2-8°C) for best results.
- Room temp is okay (for a while): They can last at room temperature (15-25°C) for up to 12 weeks without issues.
- Everything else: Store other kit components dry at room temperature (15-25°C), good for at least 18 months.
- Important: If storing everything cold, remember to warm up the buffers before use.
