Principle
- Sample lysis and digestion facilitated by lysate and Protease.
- DNA is released into the lysate.
- DNA adsorption onto magnetic particles’ surface, excluding impurities like proteins.
Procedure:
- Sample lysis and digestion: Lysate and Protease act on the sample.
- DNA release: DNA is released into the lysate.
- DNA adsorption: Magnetic particles and binding solution are added, facilitating DNA adsorption.
- Impurity removal: Proteins and impurities are removed without DNA adsorption.
- Washing:
- First, wash with washing solution to remove proteins and impurities.
- Ethanol wash to eliminate salts.
- Elution: DNA is eluted using an Elution Buffer.
Advantages:
- High specificity: Selective DNA binding, excluding impurities.
- Efficiency: Rapid purification process.
- Minimal sample loss: Enhanced DNA recovery.
Applications:
- Ideal for PCR and Southern blotting experiments requiring purified DNA.
- Suitable for various sample types and DNA sources.
Advantages
- High Binding Force: Magnetic beads with exceptional binding capacity efficiently capture DNA molecules even from samples with abundant DNA, such as whole blood, buffy coat, and concentrated blood.
- Rapid Processing: The use of polydisperse magnetic beads ensures a fast magnetic response, leading to significantly reduced extraction times.
- High-Purity DNA: The optimized protocol delivers highly purified DNA, eliminating the need for further purification steps. This high-quality DNA is directly usable for various downstream applications:
- Second-generation sequencing (NGS)
- PCR-based detection methods
- Gene bank construction
- And other DNA analyses requiring high purity

Storage and Stability
Proteinase K, MagPureParticles should be stored at 2-8°C upon arrival. However, short-term storage(up to 24 weeks) at room temperature (15-25°C) does not affect their performance. The remaining kit components can be stored at room temperature (15-25°C)and are stable for at least 18 months under these conditions.
