Introduction

Diff-Quik Stain is a rapid staining method based on Wright Stain, which is commonly used in cytology. The staining solution is prepared using the rapid staining method recommended by the World Health Organization (WHO). Similar to Wright Stain, it is based on the principles of Romanowsky Stain technology. Diff-Quik Stain yields results similar to Wright’s, but with a shorter staining time, usually within 90 seconds.

Diff-Quik Stain is primarily used for blood cell smears, bone marrow smears, vaginal secretion smears, and exfoliated cell smears. It provides batch staining with a clear background and no sediment.

Protocols (For reference only)

  1. Prepare blood smear or bone marrow smear using routine methods and fix in methanol for 20 seconds after natural drying.
  2. Stain with Diff-Quik I for 5-10 seconds (lifting the section up and down 2-3 times to distribute the stain evenly), then immediately remove.
  3. Stain with Diff-Quik II for 10-20 seconds (lifting the section up and down 2-3 times to distribute the stain evenly), then immediately remove.
  4. Observe under the microscope immediately while still wet after washing in water, or after dehydration in 95% ethanol for 5 seconds, 100% ethanol (I) for 5 seconds, 100% ethanol (II) for 30 seconds, followed by transparency achieved by xylene for 1 minute, and finally sealing.

  • Matrix, Lymphocyte: Purple
  • Cytoplasm, Red Blood Cell: Pink Red

Note

  1. Ensure blood smear or bone marrow smear thickness is uniform to avoid staining effects.
  2. Blood cell smear staining requires fresh whole blood or EDTA anticoagulant.
  3. Bone marrow smear staining requires rapid air drying to prevent cell shrinkage or deformation due to air humidity; avoid drying with high temperature or fire.
  4. Vaginal secretion smear staining requires immediate fixation with flame or alcohol to avoid cell deformation.
  5. After natural drying, fix with stain to prevent flaking.
  6. Do not directly remove or wash the smear; do not pour out the stain first to prevent settling on the smear.
  7. The stain can be reused, but filter if sediment occurs before reuse.
  8. Decolorize with methanol or alcohol if the stain is too dark; avoid re-staining.
  9. Adjust staining time or concentration if the stain is too dark or too light.
  10. Ensure the section is clean and free of acid and alkali pollution to avoid affecting staining effects.
