IntroductionTrypan Blue Stain Solution, 0.4%, an azo dye derived from toluidine, also known as diamine blue or Niagara blue, serves as a vital stain in distinguishing viable cells from non-viable ones. It selectively stains dead tissues or cells blue, leaving live cells or tissues with intact cell membranes uncolored. The solution does not absorb into viable cells due to their selective permeability. However, it penetrates the cell membrane of dead cells, causing them to appear blue under the microscope. This method, known as dye exclusion, is commonly employed in microscopy for cell counting.
Protocol
  • Collecting Cells: Centrifuge an aliquot of the cell suspension at 1000-2000 rpm for 1 minute and discard the supernatant. Resuspend the cell pellet in PBS.
  • Trypan Blue Stain: Mix 1 part Trypan Blue Stain Solution, 0.4%, with 9 parts of the cell suspension (cell dilution). Incubate the mixture for approximately 3 minutes at room temperature.
  • Cell Count: Transfer a drop of the trypan blue/cell mixture to a hemocytometer. Place the hemocytometer on the stage of a binocular microscope and focus on the cells. Viable cells remain unstained, while non-viable cells will be stained blue. Count the stained and unstained cells separately in the hemocytometer.
Note– Total number of viable cells per mL of aliquot = (Number of viable cells / Total number of cells) x 100. – Avoid prolonged staining time to prevent dye accumulation on living cells, which may affect detection results. – If precipitation occurs in the staining solution before use, filter it out and reuse it. – Exercise caution as there is a potential cancer risk associated with the substance. For safety and health, wear laboratory attire and disposable gloves during handling.
